ABSTRACT

Abstract: Olive is grown in semi-arid climatic conditions; however, little is known about mineral changes in olive plant and nutrient requirements during the production period. Hence, the current study was conducted under Pothwar agro-climatic conditions in order to select appropriate stage of macronutrients (N, P, K) application in relation to soil and leaf nutritional status during 2017 and 2018 growing seasons. Soil and leaf analysis were performed at four different phenological stages (i.e. flowering, fruit setting, fruit enlargement and fruit maturity stages). The results revealed that the assessed macronutrient in leaf and soil varied significantly among varieties, phenological stages and growing year. The results revealed also that nitrogen level was found to decrease from fruit set (1.56%) to fruit enlargement stage (1.47%). Leaf and soil N, P and K contents were found higher before the flowering (stage 1) and depleted after fruit harvesting (stage 4), regardless of olive varieties. However, high yielding varieties showed lower nutrients after fruit harvesting (stage 4). Therefore, N content in leaf and soil gradually decreased during fruit growth and development. Whereas, K content in leaf and soil sharply declined from fruit maturity to fruit ripening stage. Overall, the trend of nutrient depletion showed that plants need phosphorus for fruit setting, nitrogen before and after fruit setting, and potash after pit hardening or at oil accumulation stages.

ABSTRACT

The ethanolic extract of resinous sediment (EERS) of Etlingera elatior young inflorescence was examined for its anticancer effect and potential antioxidant activity. The anticancer effect of the EERS was evaluated on four human cancer cell lines, HCT 116, HT-29, Hela, and MCF-7, using the MTT assay. GC-MS analysis showed that the main components found in the EERS were nonyl cyclopropane (4.44%), 1-tetradecane (3.66%), cyclotetradecane (2.41%), cyclododecane (1.92%), and 1-decene (1.72%). The antioxidant activity was determined through different methods. High amounts of TPC and TFC in the EERS were found. Moderate antioxidant capacity of the EERS was detected by DPPH and ABTS assays, with EC50 values of 44.19 and 56.61 µg/mL and a high FRAP value of 281.79 nmol Fe+2 equivalent/mg extract. In the MTT assay, the EERS showed potent anticancer activity, with IC50 values of 19.82, 37.001, 50.49, and 53.29 µg/mL against HT-29, HCT 116, Hela, and MCF-7 tumour cell lines, respectively. Moreover, the results were comparable to or less potent than the standard reference drug, 5-fluorouracil. The results showed that the EERS of Etlingera elatior inflorescence contained a high amount of polyphenols and flavonoids, which may to the selective antiproliferative effects towards colon cancer in vitro

ABSTRACT

BACKGROUND: Piercing/sucking insect pests in the order Hemiptera causes substantial crop losses by removing photoassimilates and transmitting viruses to their host plants. Cloning and heterologous expression of plantderived insect resistance genes is a promising approach to control aphids and other sap-sucking insect pests. While expression from the constitutive 35S promoter provides broad protection, the phloem-specific rolC promoter provides better defense against sap sucking insects. The selection of plant-derived insect resistance genes for expression in crop species will minimize bio-safety concerns. RESULTS: Pinellia ternata leaf agglutinin gene (pta), encodes an insecticidal lectin, was isolated and cloned under the 35S and rolC promoters in the pGA482 plant transformation vector for Agrobacterium-mediated tobacco transformation. Integration and expression of the transgene was validated by Southern blotting and qRT-PCR, respectively. Insect bioassays data of transgenic tobacco plants showed that expression of pta under rolC promoter caused 100% aphid mortality and reduced aphid fecundity up to 70% in transgenic tobacco line LRP9. These results highlight the better effectivity of pta under rolC promoter to control phloem feeders, aphids. CONCLUSIONS: These findings suggested the potential of PTA against aphids and other sap sucking insect pests. Evaluation of gene in tobacco under two different promoters; 35S constitutive promoter and rolC phloemspecific promoter could be successfully use for other crop plants particularly in cotton. Development of transgenic cotton plants using plant-derived insecticidal, PTA, would be key step towards commercialization of environmentally safe insect-resistant crops.

ABSTRACT

Objective: To evaluate the anti-angiogenic and anticancer activities of Psidium guajava leaf extracts against angiogenesis-dependent colorectal cancer. Methods: Three extracts were produced using distilled water, ethanol, and n-hexane as solvents. The extracts were physically characterised through gas chromatography-mass spectrometry, ultraviolet-visible spectroscopy, and Fourier transform infrared spectroscopy. Their antioxidant activity was evaluated using the 2,2-diphenyl-1-picrylhydrazyl, total phenolic content, and total flavonoid content assays. To assess their anti-angiogenic activity, cell viability and rat aortic ring assays were conducted, while cell migration, tube formation, colony formation, and VEGF ELISA assays were conducted to elucidate their effects on different aspects of angiogenesis. Molecular docking was used to assess the anti-angiogenic potential of some possible compounds in the extracts. Tumour spheroid assay was used to assess the extracts' potential as a treatment for colorectal cancer. Results: The ethanol extract showed the best antioxidant activity. The distilled water and ethanol extracts exhibited more inhibitory activity against EA.hy926 cell viability and aortic ring microvessel growth. In addition, the ethanol extract performed significantly better than the distilled water extract against cell migration and colony formation, and VEGF expression of the cells was suppressed by the ethanol extract. Both the distilled water and ethanol extracts showed significant inhibitory effect on EA.hy926 tube formation and tumour spheroids consisting of EA.hy926 and HCT116 cells. The ethanol extract contained β-caryophyllene and β-elemene by phytochemical analysis and subsequent docking studies, which may contribute to its anti-angiogenic activity. Conclusions: The ethanol extract of Psidium guajava has potential in the treatment of colorectal cancer through the inhibition of angiogenesis.

Background: Diabetic wound is a major socioeconomic debilitating problem in this society. Various treatment options are available but still it requires better treatment option. In diabetes mellitus the oxygenation to the tissues is reduced. In this study effects of low level laser therapy were compared with topical application of Streptococcus thermophilus on diabetic wounds that induces formation of new blood vessel and free radical scavenging system, a comparative study to get better treatment option for diabetic wounds.Methods: 18 male rats were selected and divided randomly into three groups. Diabetes was induced in all the rats by using the Alloxan monohydrate at a dose of 120mg/kg of the body weight. Group A was treated with normal saline, group B was treated with low level laser therapy and group C was treated with Streptococcus thermophilus topically. Skin tissues were collected on day three and seven, slides were prepared for microscopic examination to observe the new blood vessels formation.Results: Mean number of new blood vessel formation was observed in group B compared with group A and C. Significant vasculogenesis was seen in group B when treated with Low level laser therapy.Conclusions: In the group of low level laser therapy new blood vessel formation was seen with better wound healing. It means LLLT provides better oxygenation to the tissues by generation of new blood vessels compared with Streptococcus thermophilus and normal saline.

ABSTRACT

The current study evaluated antidiabetic and antihyperlipidemic activities of aqueous extract of Sigesbeckia orientalis L. (St. Paul's Wort) (AESO) in an alloxan-induced diabetic rat model. Following OECD guidelines safe doses of AESO were assessed in rats for the main study. Serum/bood glucose, α-amylase, and lipids levels and histopathological evaluations were conducted to assess antidiabetic and associated antihyperlipidemic efficacies of AESO. AESO was found to be safe up to the dose of 2000 mg/kg. Significant (p < 0.05) reduction in glucose and lipids (total cholesterol, triglycerides, low-density lipoproteins) levels was observed in AESO treatment groups. Serum α-amylase, high-density lipoproteins, and total body weight was increased significantly (p < 0.05) in diabetic rats treated with AESO. Histopathological data showed improvement in hepatocyte and pancreatic β-cells islets architecture. HPLC analysis identified quercetin, gallic acid, vanillic acid, p-coumaric acid, m-coumaric acid and cinnamic acid in AESO which are suggested to be responsible for observed antihyperglycemic and antihyperlipidemic attributes. Further studies to standardise the extract and evaluation of safety profile in long-term toxicity studies are recommended for safe and effective antidiabetic nutraceuticals development.

ABSTRACT

Essential thrombocytosis [ET] has rarely been reported with autoimmune rheumatic disorders. We report a case of young female, diagnosed case of Rheumatoid arthritis [RA], who had been overlooked for her raised platelet counts. Later her symptoms of impending digital gangrene led to an active search for her thrombocytosis. JAK2 mutation came out to be positive and she was diagnosed as ET associated with RA. She was treated with Hydroxyurea and Aspirin, in addition to her RA treatment. Patient responded well to the treatment and her platelet counts have been gradually improved, however, she developed gangrene of toe, for which amputation of distal phalanx of toe and nail excision was done, later in the disease course

Aims: To identify strengthens and weaknesses of Tuberculosis (TB) surveillance system of District Hyderabad and suggest recommendations for improvement. Study Design: Descriptive evaluative study Place and Duration of Study: This research work was conducted in Provincial directorate of health, district health offices, and TB sentinel sites of District Hyderabad, between February and March, 2012. Methodology: A total of 26 stakeholders were identified by using purposive sampling technique based on their involvement in and relevance to the TB surveillance system. Data was collected by review of medical records and stakeholder interviews by using “Centers of Disease Control (CDCs) Updated Guidelines for the Evaluation of Surveillance Systems. Attributes were rated as good, fair and poor on the basis of obtained score. Results: System was good in simplicity and flexibility due to simple case definition and adaptation of new changes. Presence of laboratory component and 90% completeness of forms led to good rank in data quality. The system covered limited (n=12) health facilities which caused fair representativeness. The staff was highly motivated to provide accurate, consistent and complete information and suggested good acceptability of system. Due to quarterly reporting, timeliness was poor and led to delays in outbreak identification and mitigation responses. Sensitivity and positive predictive value were 26% and 56.8% respectively. The system was fair in stability as supported by ministry of health with multinational donors. Conclusion: The TB Surveillance system was satisfactory in all attributes except representativeness, sensitivity, and timeliness. Increased establishment of sentinel sites at public and private health facilities and added frequency in reporting time were recommended.

The work on maize doubled haploid development has started at the Maize Research Station, Ayub Agricultural Research Institute, Faisalabad, Pakistan during 2010 in collaboration with University of Agriculture Faisalabad, Pakistan. The aim of current study was to develop locally adopted maize haploid inducer lines utilizing cheap and easy source of Stock6 and indeterminate gametophyte mutant ig1 gene lines imported from Maize genetic Coop Stock Centre. The lines were later utilized to produce doubled haploid inbred lines. The environmental conditions of Faisalabad are extreme in nature. In spring crop during pollination period temperature may reach up to 45°C. Therefore, locally adopted haploid inducers are needed. Best performing local inbred lines were screened having branched heavy tassel, bold seed, good pollen shedding ability and vigor. These lines were utilized as female donor parent while imported lines as recurrent pollen parent in back cross breeding program until BC4 generation. These lines were selfed twice until BC4F2 and evaluated for haploid induction rate (HIR). Lines with high HIR were further intercrossed to achieve maximum transgressive segregation. Mass selection for the adaptation traits was exercised for individual F2 plants followed by ear-to-row plantation of selected progeny. Four best haploid inducer lines with HIR up to 5% having very good tassel size, height and heat stress tolerance were selected in 2014. Haploid seeds collected in different induction crosses until 2012, were used for colchicine doubling treatment. The doubling percentage of the haploid plants was very low 0.15% and out of 1000 treated seedling 15 survived and only 5 reached up to maturity, where only one D0 cob was harvested. This was successfully selfed and was grown in three different locations for next two seasons and there was no segregation in the successive generations. Developing countries that cannot afford costly haploid inducer lines can work on Stock6 and other cheap sources available free of cost and can develop their own haploid inducer lines well adapted to their own climatic conditions.

A multi-centred study was designed to collect dengue epidemiologic data from government and registered private hospitals/clinics and maintained archive of frozen specimens in bio-bank to be used for future dengue epidemic control program, and assess the epidemiology of dengue fever (DF) by evaluating biochemical and oxidative status of patients. ELISA IgM antibodies test was done to confirm DF. From August 2010 to December 2011, 101 confirmed blood samples of DF patients referred to pathology lab of Jinnah Hospital Lahore were subjected to the epidemiologic assessment by evaluating the biochemical and physiological indices and alterations of circulating antioxidants. Clinical features of DF patients and effect of fever on blood components and serum proteins of liver were recorded. The hospital stay in DF, dengue hemorrhagic fever (DHF) and dengue shock syndrome (DSS) showed significant difference. Significant increases in serum alanine amino transferase (ALT) (P=0.000), aspartate amino transferase (AST) (P=0.000), alkaline phosphatase (ALP) (P=0.000), malondialdehyde (MDA) along with significant decreases in total protein (TP) (P=0.000), reduced glutathione (GSH) (P=0.000), superoxide dismutase (SOD), catalase (CAT) (P=0.000), and sialic acid contents (P=0.016) were observed. A positive correlation existed between bound sialic acid levels, liver enzymes and circulating antioxidants (r=0.656, P=0.016). In the present study, alterations of circulating antioxidants in DF suggest that DF might be a metabolic response to an acute, self-limiting tropical viral infection, and a consequence of the viral inflammatory process.

Trachyspermum ammi Linn. (T. ammi) is an aromatic, grassy, annual plant belonging to Umbelliferae family which grows in the east of India, Pakistan, Iran, and Egypt. T. ammi has been used traditionally to treat arthritis, colic, diarrhea and gastrointestinal problems. In addition to these medicinal uses, T. ammi continues to be valued around the world as an important cooking spice and is believed to relief the common cold, flu-like symptoms, headaches, and even painful menstrual periods. These multiple uses can be explained by its several active compounds. The phytochemical studies on T. ammi seeds have revealed the presence of alkaloids, steroids, fixed oils, glycosides, tannins, saponin and flavonoids, cumene, thymene, amino acids and dietary fiber essential oils like thymol, c-terpinene, p-cymene. Several pharmacological studies on anti-tussive effect, inhibitory effect on histamine (H1) receptors, antihypertensive, antispasmodic, bronchodilator, hepato-protective, analgesic, anti-inflammatory, antioxidant and anti mutagenic activities of T. ammi seed extracts have been reported in the literature. The present review is therefore, an effort to give a detailed survey of the literature on traditional, phytochemical and pharmacological properties of T. ammi.

ABSTRACT

The objective of this study was to assess the improvement in knowledge and attitude of Pharmacy student towards diabetes by implementing one week long education program. Study design: Study was conducted between April 25, 2011 to April 29, 2011 which engaged the 3rd professional year Pharm. D. students comprising two groups: experimental and control groups. Lectures and visual presentations on diabetic education and practice were conducted for five days. Three surveys were performed on two groups i.e. non intervened control, non- intervened experimental and intervened experimental surveys. Questionnaire was used as survey tool comprising of 46 questions under ten categories. Results: Scores in all diabetic knowledge aspect was increased after education intervention and was significantly (P<0.05) different from that of control group. Conclusion: The marked increase in knowledge about diabetes among pharmacy students illustrates the importance of educational intervention. Educational programs may markedly increase the educational skills, efficiency and confidence of pharmacy students as well as professionals.


El objetivo del estudio fue evaluar cómo mejorar el conocimiento y la actitud de los estudiantes de Farmacia hacia la diabetes, mediante la implementación de un programa de educación de una semana. Diseño: El estudio se realizó entre el 25 y el 29 de abril de 2011, entre estudiantes de 3er año de la carrera de Farmacia, conformados en dos grupos: uno experimental y otro de control. Se llevaron a cabo conferencias y presentaciones visuales en educación y práctica de la diabetes durante cinco días. Tres estudios se realizaron en dos grupos: el grupo control y experimental no intervenido y el de estudio experimental intervenido. Un cuestionario se utilizó como herramienta de estudio con 46 preguntas en diez categorías. Resultados: Las puntuaciones en el conocimiento sobre todo aspecto de la diabetes se incrementó después de la intervención educativa, lo que fue significativo (P <0,05) diferente de la de grupo de control. Conclusión: El marcado aumento en el conocimiento sobre la diabetes entre los estudiantes de farmacia ilustra la importancia de la intervención educativa. Los programas educativos pueden aumentar notablemente la capacidad educativa, la eficiencia y la confianza tanto en los estudiantes de farmacia, así como en los profesionales.

ABSTRACT

The present study was carried out to determine the prevalence of families having mental retardation in Pakistani population. We enrolled seven mentally retarded (MR) families with two or more affected individuals. Family history was taken to minimize the chances of other abnormalities. Pedigrees were drawn using the Cyrillic software (version 2.1). The structure of pedigrees shows that all the marriages are consanguineous and the families have recessive mode of inheritance. All the families were studied by linkage analysis to mental retardation locus (MRT1)/gene PRSS12. Three STR markers (D4S191, D4S2392, and D4S3024) in vicinity of mental retardation (MR) locus (MRT1)/gene PRSS12 were amplified on all the sample of each family by PCR. The PCR products were then genotyped on non denaturing polyacrylamide gel electrophoresis (PAGE). The Haplotype were constructed to determine the pattern of inheritance and also to determine that a family was linked or unlinked to gene PRSS12. One out of the seven families was potentially linked to gene PRSS12, while the other six families remain unlinked.
